The buttons mimic the drawbar presets found on the black keys at the left end of a Hammond (A100, B2, B3, C2, C3 etc). Unfortunately they require two hands to access, because you have to push the shift key to get to them.
-------
Edit: sorry, this is completely wrong. I thought you were talking about the buttons below the draw bars on the virtual drawbar Nord systems. There are two buttons below each drawbar that change the draw bar settings up and down, but when shift selected get you various presets like the black keys on a real hammond.

The Preset 1 and 2 buttons let you play each program with two different bars-settings. So a quick access in any song to a different bars setup. And the Preset 2 button can do a more thing, if you want, to "read" your live bars settings (to play with a tone generated by your actual position of bars).
(sorry for my English)

So, press the Preset 1 button and change the bars as you like. Press the Preset 2 button and change the bars in a different position. Save (Store) your program. When you will load it you can play with two different setups on the fly.
Pressing Shift-Preset 2 button will "read" your actual bars position.

Ah...now that makes sense...Thank You.
*One thing to note is if you are in split mode the Preset 1 & 2 buttons don't really act as Presets they switch between the lower & upper drawbar settings.

And if You're running a MIDI keyboard You can set split so You get the lower manual on the 5D and the upper manual on the MIDI keyboard.

On a Hammond B3 (or similar model) there are two sets of 9 drawbars for each keyboard manual. You can always have two drawbar setups created and quickly switch between them. The Preset 1 and 2 lets you do the same on the Electro or Stage. As an example, 1 could be setup for a jazz type setting (16', 5 and 1/3' and 8' out all the way). Later on in the song maybe you would like more high frequencies for a Gospel type sound (such as adding the 1 and 1/3' and 1' to the existing). The Presets allow you to have them ready to go and switch between without having to work multiple drawbars.

This organ preset thing had me puzzled too.
After reading the replies I started fiddling around.
This on the Electro 5D
To use the presets effectively they need to be stored in a Program.
They can be changed 'on the fly; from there (that Program) in performance but the won't be stored.
So you can create an example of two different settings (to emulate upper and lower keyboards on a B3) and store that example as a Program.
To make the example, select a blank program.
Then choose Organ for Upper (could be lower if you'd rather).
Then set the drawbars to the setting you'd like for the upper (B3) keyboard, along with the effects and press shift and Preset 1 together and the setting will be stored (temporarily) in Preset 1.
Now set the drawbars and effects to some other setting that you like and press store and Preset 2 (for what would be the lower B3 keyboard). Now both those presets remember the two different settings.
But they will be lost if you don't store them in the empty program you have selected. So store them there and name the program. Then you can put it into a Set List.
Feel free to correct anything you think I've got fuddled up here.
