Is the difference worth it? In my opinion, yes. The EV will give you much better sound quality than the Mackie SRM450 and will more than justify the extra 180 USD in price.
This is not to knock the Mackies - I have a pair of SRM450s and I use them regularly quite happily, but the truth is they were designed a long time ago and modern equivalents from any major manufacturer will beat them very easily in terms of sound quality.

It probably doesn't make a huge difference. The important difference in my opinion is between 8" speakers and larger sizes. 8" speakers can't carry the power and depth that larger speakers can. I also have an EV ZXA1 (8") and while it sounds lovely, I simply can't use it at larger and louder gigs because it's not loud enough. Both 10" and 12" speakers should be adequate for your purposes. Some 12" speakers may actually produce too much bass for some situations, for example it's often said that the 10" QSC is a better bet than the larger 12" version.

To be honest, listening to them is by far the most important part of making a choice. Reading can only tell you so much, you own ears will instantly tell you which is the right choice for you - and it can be surprisingly different to what you thought before you actually hear them for yourself.
